1. 5x樣品緩沖液（10ml）：0.6ml 1mol/L嘅Tris-HCl（pH6.8），5ml 50%甘油，2ml 10%嘅SDS，0.5ml巰基乙醇，1ml 1%溴酚藍(lán)，0.9ml蒸餾水?？蓡?℃保存數(shù)周，或者喺－20℃保存數(shù)月。
2.凝膠貯液：?jiǎn)胀L(fēng)櫥中，稱羅丙烯酰胺30g，甲叉對(duì)丙烯酰胺0.8g，加重蒸水溶解后，定容到100ml。隔后置棕色瓶中，4℃保存，一般可放置1個(gè)月。
3. pH8.9分離架嘛！膠緩沖液：Tris 36.3g，加1mol/L HCl 48ml，加重蒸水80ml令其溶解，調(diào)pH8.9，定容至100ml，4℃保存。
4. pH6.7濃縮膠緩沖液：Tris 5.98g，加1mol/L HCl 48ml，加重蒸水80ml令其溶解，調(diào)pH6.7，定容至100ml，4℃保存。
5. TEMED（四乙基乙二胺）原液
6.10%過硫酸銨（用重蒸水新鮮配制）
7. pH8.3 Tris-甘氨酸電極緩沖液：稱羅Tris 6.0g，甘氨酸28.8g，加蒸餾水約900ml，調(diào)pH8.3后，用蒸餾水定容至1000ml。置4℃保存，臨用前稀釋10倍。
8.考馬斯亮藍(lán)G250染色液：稱100mg考馬斯亮藍(lán)G250，溶于200ml蒸餾水中，慢慢加7.5ml 70%過氯酸，zui后補(bǔ)足水到250ml，攪拌1鐘，小孔濾紙隔。
設(shè)備實(shí)驗(yàn)

電泳儀，電泳槽，水浴手，搖床。
步驟實(shí)驗(yàn)

1.樣品制備
將蛋白質(zhì)樣品與5X樣品緩沖液（20ul＋5ul）喺一個(gè)Eppendorf理中撈。放入100℃加熱5－10min，取上清啲呀。
2.分離架嘛！膠同濃縮膠嘅制備
1）將玻璃黑、樣品呀，Spacer用洗滌劑洗凈，用ddH2O沖洗數(shù)次，而且用乙醇擦拭，晾干；
2）將兩蚊洗凈嘅玻璃板之間加Spacer，按照Bio-Rad MiniⅡ/Ⅲ說明書顯示裝好玻璃板；
3）按如下體積配制10%分離架嘛！膠8.0 ml，撈勻；
ddH2O 3.0 ml
1.0 mol/LTris-HCl pH=8.8 2.1 ml
30%Acr-Bis 2.8 ml
10%SDS 80 ul
10%AP 56 ul
TEMED 6 ul

1. 5x sample buffer (10ml): 0.6ml 1mol/L Tris-HCl (pH6.8), 5ml 50% glycerol, 2ml 10% SDS, 0.5ml sulfhydryl alcohol, 1ml 1% bromo phenol blue, and distilled water. Can be kept for several weeks at 4 degrees, or stored for a few months in from-20 deg.
2. gel liquid storage: in a hood, and acrylamide 30g, methylene bisacrylamide 0.8g, add distilled water dissolved, constant volume to 100ml. After filtering, the brown bottle is stored at 4 C, and usually can be placed for 1 months.
3. pH8.9 separation gel buffer: Tris 36.3g, 1mol/L HCl and 48ml 80ml, add distilled water to dissolve, adjust the volume to 100ml, pH8.9, 4 degrees to save.
4. pH6.7 concentrated gel buffer: Tris 5.98g, 1mol/L HCl and 48ml 80ml, add distilled water to dissolve, adjust the volume to 100ml, pH6.7, 4 degrees to save.
5. TEMED (four ethyl ethylenediamine) original solution
6.10% ammonium persulfate (with distilled fresh preparation)
7. pH8.3 Tris- glycine electrode buffer solution: Tris 6.0g, glycine 28.8g, adding distilled water about 900ml, pH8.3, with distilled water to be fixed to 1000ml. Store at 4 C and dilute 10 times before use.
8. Coomassie brilliant blue G250 staining solution: 100mg Coomassie brilliant blue G250, dissolved in 200ml distilled water, slowly adding perchloric acid 7.5ml 70%, finally make up the water to 250ml, stirring for 1 hours, pinhole filter.
Experimental equipment

Electrophoretic apparatus, electrophoretic trough, water bath pot, rocking bed.
Experimental steps

1. sample preparation
The protein sample and 5X sample buffer (20ul5ul) in a mixed Eppendorf tube. Heat at 100 C and heat 5 - 10min, Torikami Kiyo sample.
Preparation of 2. sealant and concentrate
1) the glass plate, the sample comb, the Spacer washing with detergent, ddH2O washing several times, then the ethanol wiping, dry;
2) add two pieces of clean glass plate to Spacer and install the glass plate according to the instructions of Bio-Rad Mini II / III.
3) make up 10% separate glue 8 ml according to the following volume, and mix well.

ななしち. pH8.3 Tris -グリシン電極緩衝液：とTris 6.0g取り、グリシン28.8gに加え、蒸留水は約900ml、調(diào)pH8.3後、蒸留水定容～1000 ml。4℃で保存し、使用前に10倍まで希釈する。
はち.クマシーブリリアントブルーG250染色液とクマシーブリリアントブルーG250：100 mg、200 mlの蒸留水に溶けて、徐々に加わる7.5ml 70％の過塩素酸、zui後まで補(bǔ)足水250 ml攪拌いち?xí)r間、孔ろ過ろ紙。
実験設(shè)備

水泳器、スイミングスロット、水浴鍋、床を橫に振る。
実験の手順

1 .サンプル制
サンプルは蛋白質(zhì)と5Xサンプル緩衝液（20ul＋5ul）にEppendorf管で混合。ひゃく℃を入れて加熱5－10minを點(diǎn)検するように。
