【流感AB病毒試紙 流感甲乙型病毒聯(lián)檢試紙】

（1）細菌性肺炎發(fā)生率為5～15%。流感起病后2～4天病情進一步加重，或在流感恢復期后病情反而加重，出現(xiàn)高熱、劇烈咳嗽、膿性痰、呼吸困難，肺部濕性啰音及肺實變體征。外周血白細胞總數(shù)和中性粒細胞顯著增多，以肺炎鏈球菌、金黃色葡萄球菌，尤其是耐甲氧西林金黃色葡萄球菌，肺炎鏈球菌或流感嗜血桿菌等為主。

（2）其他病原菌感染所致肺炎包括衣原體、支原體、嗜肺軍團菌、真菌（曲霉菌）等，對流感患者的肺炎經(jīng)常規(guī)抗感染治療無效時，應考慮到真菌感染的可能。

（3）其他病毒性肺炎常見的有鼻病毒、冠狀病毒、呼吸道合胞病毒、副流感病毒等，在慢性阻塞性肺部疾病患者中發(fā)生率高，并可使病情加重，臨床上難以和流感病毒引起的肺炎相區(qū)別，相關病原學和血清學檢測有助于鑒別診斷。

（4）Reye綜合征（瑞氏綜合征）偶見于14歲以下的兒童，尤其是使用阿司匹林等水楊酸類解熱鎮(zhèn)痛藥物者。主要表現(xiàn)為退熱后出現(xiàn)嘔吐、繼之嗜睡、昏迷、驚厥等神經(jīng)系統(tǒng)癥狀，肝大，無黃疸，腦脊液檢查正常。發(fā)病機制不清楚。

（5）心臟損害心臟損傷不常見，主要有心肌炎、心包炎?？梢娂∷峒っ干?、心電圖異常，而肌鈣蛋白異常少見，多可恢復。重癥病例可出現(xiàn)心力衰竭。

（6）神經(jīng)系統(tǒng)損傷包括腦脊髓炎、橫斷性脊髓炎、無菌性腦膜炎、局灶性神經(jīng)功能紊亂、急性的感染性脫髓鞘性多發(fā)性神經(jīng)根神經(jīng)?。ǜ窳职屠C合征）。

（7）肌炎和橫紋肌溶解綜合征在流感中罕見。主要癥狀有肌無力、腎衰竭，CK升高。

包涵體的復性還要注意以下幾點： 
1.首先要獲得較高純度的包涵體。 
2.包涵體溶解要*，一般應使溶解液體量較大，不要怕蛋白被稀釋，要有助溶措施。 
3.透析前后均要離心 
4.透析不能太快. 
5.純化的條件要摸索。 
小技巧： 
1）包含體要*洗滌干凈，洗滌液中加入適量Triton-X100. 
2) 包含體溶解后裝入透析袋在復性液中復性 
3）復性液的組成很有講究，本人使用 Oxidised Glutathione/ Reduced Glutathione 還加入一定的L-Arginine-Hydrochloride 12-24h 
4) 復性完畢在低濃度的緩沖液中連續(xù)緩慢透析12-24h 
5) 復性率的測定 據(jù)變性蛋白和非變性蛋白的光學性質不同測定（望有經(jīng)驗的戰(zhàn)友能更詳細地講解）  
6）TritionX100、SDS這一類去垢劑zui后不易去除，在制藥行業(yè)中一般不允許采用。好像SDSzui后殘留量不能大于0.5%吧，忘了。 
我用Triton洗滌有些包涵體效果不是很好。包涵體洗滌是純化極為重要的一步，改變培養(yǎng)條件，再洗滌包涵體，有時可以在上柱前已經(jīng)只剩下3-4條雜帶，這樣后續(xù)的純化就很方便了。因為色譜柱的純化條件比較難optimize。

I use Triton washing some of the inclusion body effect is not very good. Inclusion body washing is an extremely important step in purification.
