【預(yù)期用途】

用于待測致敏紅細(xì)胞樣本的預(yù)處理，使致敏紅細(xì)胞樣本中的待測抗體從與細(xì)胞結(jié)合的狀態(tài)中解離釋放出來。以便于使用體外診斷試劑或儀器對待測抗體進(jìn)行檢測。

【檢驗原理】

紅細(xì)胞上的抗原與血清中抗體在適合條件下發(fā)生致敏，這種結(jié)合在一定條件下是可逆的。將已致敏的紅細(xì)胞懸浮于低pH值的甘氨酸溶液中，抗體蛋白又可以從結(jié)合的紅細(xì)胞上解離釋放出來。離心取上清解離的放散液，此液中含有從紅細(xì)胞表面解離釋放出來的抗體蛋白，經(jīng)Tris緩沖液調(diào)節(jié)pH至中性后此上清放散液可用于相關(guān)抗體的檢測和鑒定；而解離釋放后的紅細(xì)胞經(jīng)洗滌后可用于血型定型、自身抗體的吸收等。

【主要組成成份】

1.溶液I：主要組分為甘氨酸(C₂H₅NO₂)，氯化鈉(NaCl)。

2.溶液II：主要組分為Tris堿，指示劑。   

【儲存條件及有效期】                  

常溫（10～30℃）儲存，有效期1年。試劑開瓶后在常溫（10～30℃）條件下可儲存6個月。

【樣本要求】

新鮮或2～8℃保存不超過72小時的抗凝血樣。

是硬腦膜內(nèi)層自顱頂正中線折病毒并向伸展與兩半球之間的結(jié)構(gòu)。其前端窄，附于雞冠，后分寬，向下連于小腦幕的上面。
小腦幕
呈半月形，水平地位于大腦半球與小腦之間。小腦幕分為兩側(cè)顳骨巖部；前緣游離并向后凹陷，稱為幕切跡，與蝶骨鞍背圍成的孔有中腦穿過。小腦幕將顱腔分為幕上、下間隙。幕上間隙又借大腦鐮分為左、右兩部。當(dāng)幕上間隙的顱內(nèi)壓增高時，可將海馬旁回和海馬旁回鉤推入小腦幕孔，形成顳葉鉤回疝。
小腦鐮
后部附著與枕內(nèi)嵴，前緣游離，呈鐮刀狀，部分的分割小腦兩半球。向上連于小腦幕，下接枕骨大孔邊緣。
鞍隔
為環(huán)狀鄒襞，中央有一孔，漏斗從此通過。其前方附著于鞍結(jié)節(jié)和前床突，后方附著于小腦幕游離緣，構(gòu)成垂體窩的頂。
硬膜竇是由硬腦膜的骨膜層在特定部位互相分離而形成的腔隙，在腔隙內(nèi)面襯有內(nèi)皮細(xì)胞。
硬膜竇中，充以靜脈血并于靜脈相續(xù)，故稱靜脈竇。病毒其壁厚不易塌陷，當(dāng)損傷時則出血兇猛。
上矢狀竇（superior sagittal sinus）：位于顱頂中線偏右，居大腦鐮的上緣。前起盲孔，后至竇匯。主要接受大腦背外側(cè)面上部和部分內(nèi)側(cè)面的靜脈血。上矢狀竇兩側(cè)壁上有許多靜脈陷窩，蛛網(wǎng)膜絨毛伸入其中。腦脊液通過上述絨毛的再吸收作用而進(jìn)入靜脈竇。病毒此，上矢狀竇是腦皮層靜脈和腦脊液回流的必經(jīng)之路。
下矢狀竇（inferior sagittal sinus）：位于大腦鐮下部的游離緣，在小腦幕的前緣處與大腦大靜脈匯合共同延為直竇。
直竇（rectus sinus）：位于大腦鐮和小腦幕的匯合處，直行向后，在枕內(nèi)隆凸附近與上矢狀竇匯合成為竇匯，并向兩側(cè)延伸為橫竇。
橫竇和乙狀竇：橫竇位于枕骨橫溝處，即小腦幕的后外側(cè)緣，向前行至巖枕裂處轉(zhuǎn)向下成為乙狀竇。乙狀竇位于顳骨的乙狀溝內(nèi)。
竇匯：為上矢狀竇、下矢狀竇、直竇和左、右橫竇的會合處。實際上以上各竇會合在竇匯者少見。若上矢狀竇分支時，則右支常比左支大，右橫竇也比左橫竇寬大。
