由DNA、蛋白質(zhì)和RNA構(gòu)成的細(xì)菌染色體是高度濃縮的。它不僅通過拓?fù)洚悩?gòu)酶（topoisomerase）形成超螺旋，并且，環(huán)繞在由RNA和蛋白質(zhì)形成的“核”的內(nèi)外。許多DNA的負(fù)電荷被多胺[如精胺（spermine）和亞精胺（spermidine）]和DNA纏繞著的堿性蛋白質(zhì)所中和。通過柔和地裂解細(xì)菌細(xì)胞得到的DNA外觀呈串珠狀。雖然細(xì)菌染色體也是高度濃縮的，但是，在光學(xué)顯微鏡下它們不能被看到。在透射電子顯微鏡下，細(xì)菌染色體的外觀與非分裂的真核細(xì)胞核內(nèi)的染色質(zhì)（chiromatin）非常相像。
