檢驗(yàn)原理 德國維賽維潤風(fēng)疹特異性IgM ELISA試劑盒

用抗原包被微量板孔，制成固相載體。加患者血清到板孔中，其所含的抗體特異性地與固相載體中現(xiàn)存抗原結(jié)合，形成免疫復(fù)合物。除去多余物質(zhì)后，加入結(jié)合了堿性磷酸酶的IgG、IgA或IgM抗體，使之與上述免疫復(fù)合物反應(yīng)。洗板，除去多余的結(jié)合物，加入底物（對硝基苯磷酸鹽）。其與酶結(jié)合的免疫復(fù)合物反應(yīng)，產(chǎn)生有顏色產(chǎn)物，顏色強(qiáng)度與特異性抗體含量成正比。

霍亂弧菌在未經(jīng)處理的糞便中，可存活數(shù)天；在冰箱內(nèi)的牛奶、

鮮肉和魚蝦水產(chǎn)品中存活時(shí)間分別為2～4周、1周和1～3周；在室

溫下存放的新鮮蔬菜中，可存活1～5天；在砧板和布上可存活相

當(dāng)長時(shí)間；在玻璃、瓷器、塑料和金屬上存活時(shí)間不超過2天?；?/p>

亂弧菌古典生物型在外界環(huán)境中生存能力不強(qiáng)，而愛爾托生物型

抵抗力較強(qiáng)，在河水、井水、池塘水和海水中可存活1～3周，甚

至更長，有時(shí)在局部自然水中也能越冬。愛爾托生物型弧菌可粘

附于海洋甲殼類生物表面，分泌甲殼酶，分解甲殼作為營養(yǎng)而長

期存活，如愛爾托生物型弧菌被人工飼養(yǎng)的泥鰍、鱔魚吞食后，

可在其體內(nèi)生長繁殖，然后排入水中。
霍亂弧菌對熱、干燥、日光及一般消毒劑均很敏感，經(jīng)干燥2小時(shí)

或加熱55℃10分鐘即可死亡，煮沸立即死亡；對酸敏感，在正常

胃酸中僅能存活4分鐘，接觸1∶5000～1∶10000鹽酸或硫酸、1

∶2000～1∶3000細(xì)菌或1∶500000高錳酸鉀，數(shù)分鐘即被殺滅，

在0.1%漂白粉中10分鐘內(nèi)即可死亡。氯化鈉的濃度高于4%或蔗糖

濃度在5%以上的食物、香料、醋及酒等，均不利于霍亂弧菌的生

存。
發(fā)病機(jī)制
正常胃酸可殺滅霍亂弧菌，當(dāng)胃酸分泌缺乏或低下，或入侵的霍

亂弧菌數(shù)量較多，未被殺滅的弧菌就進(jìn)入小腸，在堿性腸液內(nèi)迅

速繁殖，并通過粘液對細(xì)菌的趨化吸引作用、細(xì)菌鞭毛活動(dòng)及弧

菌粘蛋白溶解酶和粘附素等的作用，粘附于小腸粘膜的上皮細(xì)胞

表面，并在此大量繁殖。
此菌產(chǎn)生強(qiáng)烈的外毒素，即霍亂腸毒素，由A亞單位和B亞單位組

成。B亞單位與該處粘膜上皮細(xì)胞表面受體——神經(jīng)節(jié)苷脂結(jié)合，

A、B兩種亞單位解離，A亞單位穿過細(xì)胞膜進(jìn)入細(xì)胞內(nèi)，激活腺苷

酸環(huán)化酶(AC)，使細(xì)胞內(nèi)三磷酸腺苷(ATP)轉(zhuǎn)化為環(huán)磷酸腺苷

(cAMP)，使細(xì)胞內(nèi)環(huán)磷酸腺苷含量提高，促使一系列

酶反應(yīng)加速進(jìn)行，導(dǎo)致空腸到回腸部腺細(xì)胞分泌功能亢進(jìn)，引起

大量液體及血漿中的鈉、鉀、氯等離子進(jìn)入腸腔，由于分泌功能
