3、“陰陽臉”著色
指組織一半著色一半無著色,形成交界清晰或不甚清晰的兩種染色結(jié)果。其成因是試劑僅覆蓋了部分組織而不是全部。如加試劑后未讓試劑流散開而集中在部分組織上。通常應(yīng)該在加完試劑后,仔細看一遍,是否有的組織未被試劑*覆蓋,如有這種情況,建議用牙簽而不是用吸頭或試劑瓶口將試劑引流開使之將組織全部覆蓋。另外,染片盒不平,切片傾斜,雖然開始試劑已全部覆蓋了組織,但后來試劑流向一邊,部分組織未被試劑覆蓋。對于這種問題,只要留心或想到了很容易發(fā)現(xiàn),也很容易解決。有時,用DAKO(或PAP)筆在組織周圍畫圈時,劃線太靠近或畫到了組織上,由于筆油的力學原理,試劑不能達到靠近劃線附近的組織。還有氣泡也可引起陰陽分明的著色,只是不著色區(qū)域是圓形,由于氣泡中含氣,試劑被推到周圍,因此,氣泡中心的組織不著色。解決辦法是滴加試劑時手法要輕,有氣泡時用牙簽捅破。
4、灶片狀著色
切片中著色區(qū)東一塊西一塊,呈灶片狀分布,出現(xiàn)這種問題的原因有:(1)裱片時水未排盡,在局部形成氣泡使組織突起,染色時試劑滲入后不易洗盡,顯色過深所致。解決辦法是,漂片盒里的氣泡應(yīng)去盡,晾片熱臺不能平放,應(yīng)有45度左右的斜度,利于水流走和蒸發(fā)。(2)壞死組織灶,組織壞死后細胞破壞、酶的釋放、蛋白游離、分解,復(fù)雜的肽鏈殘段(如Fc段)可能與一抗或/和二抗結(jié)合導(dǎo)致zui終著色。解決辦法是在選擇染色切片時應(yīng)避免選擇壞死組織較多的切片。(3)制作APES膠片時,膠的濃度太高,干燥后在玻片上留下白色小點,顯色時白色小點著色。解決辦法是按照標準的制備方法進行,即5%鹽酸酒精(5 ml鹽酸+95%酒精95 ml)浸泡玻片4小時、熱水沖洗玻片1小時、蒸餾水洗玻片1分鐘、丙酮浸泡玻片5秒鐘后空氣干燥(室溫)、2% APES(2 ml APES+98 ml丙酮)浸泡玻片5分鐘、玻片過一下丙酮(1-2秒鐘)、玻片過一下蒸餾水(1-2秒鐘)、37 度過夜干燥、室溫儲存?zhèn)溆谩H绻破^程中,因丙酮逐漸揮發(fā)而膠變濃時可適當加入一些丙酮。

The solution is to avoid the need to select more sections of necrotic tissue when choosing a stained section. (3) When making APES film, the concentration of glue is too high, leaving small white spots on the glass slides after drying, and coloring of the white dots when coloring.
