【產(chǎn)品說(shuō)明書】

【試驗(yàn)原理】
    流行性感冒是指由流感病毒引起的具有傳染力的疾病，可引發(fā)肺炎或更嚴(yán)重的并發(fā)癥。流感病毒的結(jié)構(gòu)主要包括內(nèi)部的核心和外部的病毒囊膜。流感病毒核心是由核蛋白卷曲包繞螺旋形核糖核酸(RNA)組成，核蛋白的抗原穩(wěn)定，很少發(fā)生變異，具有型特異性。根據(jù)核蛋白抗原性的不同，可把感染人的流感病毒分為甲、乙、丙三型。本產(chǎn)品是利用能夠特異性識(shí)別甲乙型流感病毒核蛋白的單克隆抗體，以免疫色譜法檢測(cè)流感病毒核蛋白的試劑盒，本試劑盒僅對(duì)樣品中的甲乙型流感病毒有反應(yīng)，對(duì)乙型及丙型流感病毒均不反應(yīng)，而且不能用于區(qū)分病毒感染的亞型。
本品是由樣品滴下部、試劑部和展開部三部分組成的檢測(cè)板，內(nèi)藏長(zhǎng)方塊狀的載體。其中，試劑部包含膠體金標(biāo)記抗甲乙型流感病毒單克隆抗體(鼠)(以下將簡(jiǎn)稱為膠體金標(biāo)記抗體A)；展開部包含固定化的抗甲乙型流感病毒單克隆抗體(鼠)(以下簡(jiǎn)稱為抗流感A抗體)及抗鼠免疫球蛋白多克隆抗體(兔)(以下簡(jiǎn)稱為抗鼠免疫球蛋白抗體)。
檢測(cè)板各部分的名稱

樣品從檢測(cè)板的樣品滴下部滴下后，膠體金標(biāo)記抗體A被溶解，與樣品中的甲乙型流感病毒抗原形成免疫復(fù)合體。免疫復(fù)合體因展開部的毛細(xì)管現(xiàn)象而移動(dòng)，被判定部〔T〕中固定化的抗流感A抗體捕捉，于是在判定部[T]因膠體金的作用形成紫紅色的條帶。本試劑盒的這個(gè)紫紅色條帶可以肉眼確定，以判斷樣品中是否存在甲乙型流感病毒。
另一方面，無(wú)論樣品中是否有甲乙型流感病毒存在，剩余的膠體金標(biāo)志抗體A繼續(xù)在展開部移動(dòng)，在判定部[C]被固定化的抗鼠免疫球蛋白抗體捕捉，因膠體金而形成紫紅色條帶。這顯示了膠體金標(biāo)志抗體的正常移動(dòng)。

【試劑組成】 

1. 流感病毒快速檢測(cè)卡40片/盒    

2. 樣本稀釋液40管/盒      

3. 棉簽40支/盒  

【操作方法】 一、樣品制備： 

1. 本檢測(cè)卡采集樣品為眼、氣管分泌物。將棉簽插入分泌物zui多的部位，輕輕搖動(dòng)棉簽，讓棉簽充分吸收分泌物。 

2. 將棉簽在稀釋液中充分?jǐn)嚢璨⒎磸?fù)擠壓試管壁，讓分泌物充分溶解到稀釋液中，得到待檢樣品。 

3. 樣品一般須當(dāng)即進(jìn)行檢測(cè)，否則應(yīng)冷藏保存，超過(guò)24小時(shí)的，應(yīng)該冷凍保存。  

二、操作步驟： 

1. 使用前將試劑盒和樣品恢復(fù)至室溫。 

2. 將棉簽浸入裝有樣品稀釋液的試管，充分?jǐn)嚢杌靹蚝?，用一次性滴管取上清液?/span>

3. 取出檢測(cè)卡，開封后平放于桌面上，從滴管中緩慢而準(zhǔn)確地逐滴加入2–3滴混合液。 

4. 加樣品液后，約30秒內(nèi)，紅色的液體從靠樣品孔的觀察窗邊緣涌出。朝另一方向流動(dòng)。 

5. 五分鐘后判斷結(jié)果，超過(guò)三十分鐘的結(jié)果判讀無(wú)效。  

【結(jié)果判定】        

1. 陽(yáng)性：在觀察孔內(nèi)，若對(duì)照線顯色，檢測(cè)線同時(shí)顯色，判為陽(yáng)性(如圖一、圖二)。陽(yáng)性結(jié)果表明檢測(cè)液

中含有流感病毒。 

2. 陰性：在觀察孔內(nèi)，若對(duì)照線顯色，而檢測(cè)線不顯色，判為陰性（如圖三）。陰性結(jié)果表明檢測(cè)液中不含流感病毒。 

3. 無(wú)效：在觀察孔內(nèi)，若對(duì)照線不顯色，則結(jié)果無(wú)效，建議重新測(cè)試(如圖四、圖五)。   

【注意事項(xiàng)】 

1. 本品如果購(gòu)買時(shí)發(fā)現(xiàn)過(guò)期、破損、污染、無(wú)效的產(chǎn)品，請(qǐng)?jiān)谫?gòu)買處進(jìn)行更換。  

2. 測(cè)試樣品來(lái)自動(dòng)物，可能有潛在感染性，樣品和使用過(guò)的試劑應(yīng)被看作微生物危險(xiǎn)品處理。

3. 所有檢測(cè)卡啟封后馬上使用，此前不要隨意打開。 

4. 本品為一次性產(chǎn)品，請(qǐng)勿二次使用；請(qǐng)勿使用非本品隨附的稀釋液。  

【儲(chǔ)存及有效期】 

1. 保存于干燥陰涼處（2~30℃），有效期為18個(gè)月，生產(chǎn)日期見(jiàn)外包裝盒。

2. 打開包裝后，在1小時(shí)內(nèi)使用。

內(nèi)分泌系統(tǒng)的改變有三種
①通過(guò)下丘腦－垂體－腎上腺皮質(zhì)系統(tǒng)的活動(dòng)，分泌促腎上腺皮質(zhì)激素（ACTH）、抗利尿激素 (ADH)及生長(zhǎng)激素(GH)。促腎上腺皮質(zhì)激素使腎上腺皮質(zhì)分泌皮質(zhì)醇。皮質(zhì)醇參與機(jī)體能源的動(dòng)用，促進(jìn)葡萄糖異生，使血糖升高，促進(jìn)脂肪分解，產(chǎn)生能量。皮質(zhì)醇參與兒茶酚胺對(duì)血管功能的調(diào)節(jié)，幫助維持血壓。皮質(zhì)醇還能抑制炎性反應(yīng)，減少血管滲出,減輕炎癥的損害作用。創(chuàng)傷后,增加皮質(zhì)醇的分泌是身體必需的防御反應(yīng)。若無(wú)足夠量的皮質(zhì)醇可發(fā)生循環(huán)衰竭而死亡。抗利尿激素可減少水分排泄，加強(qiáng)腎遠(yuǎn)曲小管和集合小管對(duì)水分的重吸收，從而有利于維持體液容量及循環(huán)血量。
②創(chuàng)傷引起交感神經(jīng)－腎上腺髓質(zhì)的變化，分泌大量去甲腎上腺素和腎上腺素（兒茶酚胺）。創(chuàng)傷后的兒茶酚胺分泌可以調(diào)節(jié)心血管功能，保證心腦等重要臟器的血供應(yīng)，可促進(jìn)肝臟和肌肉的糖原分解，抑制胰島素分泌，同時(shí)增加高糖素，使血糖升高，可以激活脂肪酶，從而促進(jìn)貯存脂肪水解為脂肪酸，而成為主要的能量來(lái)源；同時(shí)還使肌肉釋放氨基酸。
